The Multi-Omics technology delivered by BD is designed to provide an end-to-end, complete workflow for the analysis of single cells. From sample preparation to data analysis, this unique workflow increases experimental capabilities for researchers by enabling protein profiling in addition to gene expression analysis in isolated single cells. Protein expression can be estimated using conjugated oligonucleotide barcodes (BD™ Single-cell Multiplexing Kit) or/and BD™ AbSeq oligo-conjugated antibodies to detect cell surface proteins, for a true ‘multi-omics’ assay. The use of the BD™ Single-cell Multiplexing kit enables the addition of multiple samples in a single run, thereby saving on time and reagent costs, while the BD™ AbSeq allows simultaneous determination of protein and mRNA expression. The single cell capture, as well as the molecular barcoding, are processed using the BD Rhapsody™ system, followed by library preparation, sequencing and data analysis using SeqGeq™. Several options are offered for the library preparation, from Whole Transcriptome Amplification (WTA) to targeted and custom panels.
The workflow lends itself to numerous applications, from WTA and the discovery of unique protein biomarkers to the detection of diverse clonotypes, protein expression profiling and evaluation of mRNA expression in a single experiment.
