Simultaneous analysis of mRNA and proteins in mouse immune cells using the BD™ Mouse Single-cell multiplexing kit and the BD™ AbSeq assay on the BD Rhapsody™ system

The authors of this poster have studied the effects of obesity on immune cell populations in mice. They used cells isolated from epididymal fat, thymus, spleen and bone marrow from mice placed on high fat (HFD) and control diets. DNA-barcoded universal antibodies from the BD™ Mouse Single-cell Multiplexing kit allowed them to combine 8 samples into a single pool, thereby reducing experimental scale and costs. The use of targeted immune-focused mRNA and the BD™ AbSeq antibodies provided effective unbiased clustering of immune cell populations from the mouse tissues. This helped identify four clusters in which consistent T-distributed stochastic neighbor embedding (t-SNE) differences in gene expression were seen between the HFD and normal mice. The study shows that adipose tissue macrophages (ATM) and CD11c+ myeloid cells exhibit inflammatory and adipogenic gene signatures in HFD mice. In addition, cytotoxic CD8+ cells in the HFD mice are seen co-expressing exhaustion markers. Thus, multi-omic analysis enables the identification of obesity-related phenotypes and allows the study of the underlying mechanisms behind them.
